产品简介

本产品适用于新鲜植物样本的细胞核分离。通过裂解细胞膜,释放细胞核的同时保持细胞核膜的稳定,经密度梯度离心快速地从植物 样本中分离纯化细胞核,并减少碎片及次生代谢产物的残留。

制备的细胞核悬液可用于 1、染色质可及性分析(scATAC- seq/bulk ATAC- seq,CUT&Tag);
2、核转录组测序(snRNA-seq/ bulk RNA-seq)等相关实验。

本品可在50分钟内完成4-6个样本的细胞核分离,操作简单。

  •  优化后的密度梯度离心法可减少细胞碎片及植物次生代谢产 物的残留。
  •  获得的植物细胞核形态完整,无结团。

Product Introduction

This kit is designed for nuclear isolation from fresh plant samples. It lyses cell membranes to release intact nuclei while stabilizing the nuclear envelope. Target nuclei are rapidly purified via density-gradient centrifugation, lowering residual cellular debris and plant secondary metabolites.

The prepared nuclear suspension is applicable for: 1.Chromatin accessibility assays including scATAC‑seq, bulk ATAC‑seq and CUT&Tag;
2.Nuclear transcriptome sequencing (snRNA‑seq, bulk RNA‑seq) and other related experiments.

This product enables nuclear isolation of 4–6 samples within 50 minutes with straightforward operation.

  • The optimized density-gradient centrifugation reduces residual cellular debris and plant secondary metabolites.
  •  Isolated plant nuclei feature intact morphology and are free from aggregation.

Q: 植物样本抽核后,是否能保存至第二天再进行ATAC实验?液氮研磨与剪切区别有何区别?

A: 不建议,建议分离细胞核后0.5h内进行后续实验。液氮研磨细胞核形态较好,得率高,但部分样本碎片多;剪切背景较干净,但得率偏低。不同的物种适用处理方法不一样,建议测试后选择合适的样本处理方法。

 

Q: 枇杷果实抽核是否适用?

A: 果实类抽核效果一般,枇杷果实暂无经验,建议客户自行测试。

A: 暂无此产品。

 


Q1: Can isolated nuclei from plant samples be stored overnight before ATAC‑seq? What is the difference between liquid nitrogen grinding and mechanical shearing?

A1: Storage overnight is not recommended. Downstream ATAC assay should be performed within 0.5 hours post nuclear isolation.

Liquid nitrogen grinding yields intact nuclei with higher recovery but generates more tissue debris for certain samples; mechanical shearing results in lower background yet lower nuclear yield. Optimal pretreatment varies across plant species; pilot tests are recommended to determine the proper protocol.


Q2: Is this kit applicable for nuclei extraction from loquat fruit tissues?

A2: Nuclear isolation efficiency is generally poor for fruit tissues. We have no test data on loquat fruit; customer validation is suggested.


Q3: Do you offer protoplast isolation kit for plant samples?

A3: This product is not available at present.
